En 2011 d\u00e9j\u00e0, une \u00e9quipe de chercheurs de l\u2019Inra et de l\u2019Inserm a montr\u00e9 chez le rongeur qu\u2019une faible consommation d\u2019Om\u00e9ga 3 chez les souris augmentait leur stress (1).<\/strong> Ce ph\u00e9nom\u00e8ne serait li\u00e9 \u00e0 l\u2019alt\u00e9ration de la capacit\u00e9 du cerveau \u00e0 produire des endocannabino\u00efdes (cannabino\u00efdes endog\u00e8nes), des lipides du cerveau qui contr\u00f4lent la m\u00e9moire synaptique. Pour mieux comprendre les liens entre anxi\u00e9t\u00e9 et plasticit\u00e9 synaptique d\u00e9pendante des endocannabino\u00efdes, l\u2019\u00e9quipe de chercheurs a poursuivi ses exp\u00e9rimentations en testant diff\u00e9rents mod\u00e8les de stress comportementaux sur les rongeurs. Ces travaux, qui r\u00e9v\u00e8lent que la plasticit\u00e9 d\u00e9pendante des endocannabino\u00efdes dans le noyau accumbens est le substrat neurobiologique de l\u2019anxi\u00e9t\u00e9.<br \/>\n<\/em><\/p>\n<p><strong><span class=\"image-wrapper frame floatleft\"><img loading=\"lazy\" decoding=\"async\" class=\"alignleft\" src=\"https:\/\/www.bordeaux-neurocampus.fr\/_contents-images\/ametys-internal%253Asites\/neurosciences\/ametys-internal%253Acontents\/cell-reports-s-laye-actualite\/_metadata\/content\/_data\/Bosch-Bouju-Clementine160.jpg_222x160\" alt=\"\" width=\"160\" height=\"222\" \/><br \/>\n<\/span>Cl\u00e9mentine Bosch-Bouju chercheur post-doctoral NutriNeuro: &#8221;\u00a0Les souris ne sont pas \u00e9gales face au stress. Un constat r\u00e9alis\u00e9 par une \u00e9quipe de recherche de l\u2019Inra et de l\u2019Inserm qui, apr\u00e8s avoir soumis des rongeurs \u00e0 un stress chronique et mesur\u00e9 les sympt\u00f4mes \u00e9motionnels gr\u00e2ce \u00e0 une batterie de tests comportementaux :<\/strong> isolement, labyrinthe ou environnement anxiog\u00e8ne, a remarqu\u00e9 que certains animaux \u00e9taient naturellement r\u00e9silients, autrement dit plus r\u00e9sistants au stress. Les chercheurs ont \u00e9galement mis en lumi\u00e8re que cette capacit\u00e9 serait due \u00e0 une meilleure plasticit\u00e9 des neurones du noyau accumbens, une zone du cerveau impliqu\u00e9e dans la r\u00e9gulation des \u00e9motions et du stress, o\u00f9 les endocannabino\u00efdes sont des acteurs majeurs de la m\u00e9moire \u00e0 l\u2019\u00e9chelle synaptique.\u00a0Pour v\u00e9rifier cette relation, les souris qui pr\u00e9sentaient des sympt\u00f4mes anxieux ont re\u00e7u un traitement stimulant la production d\u2019endocannabino\u00efdes dans le noyau accumbens. Les scientifiques ont observ\u00e9 une limitation de l\u2019anxi\u00e9t\u00e9 chez ces souris. Ces r\u00e9sultats mettent en \u00e9vidence pour la premi\u00e8re fois la relation directe entre la production d\u2019endocannabino\u00efdes au niveau du noyau accumbens et le d\u00e9veloppement de troubles anxieux face \u00e0 un stress chronique.<\/p>\n<p><span class=\"image-wrapper frame floatleft\"><img loading=\"lazy\" decoding=\"async\" class=\"alignleft\" src=\"https:\/\/www.bordeaux-neurocampus.fr\/_contents-images\/ametys-internal%253Asites\/neurosciences\/ametys-internal%253Acontents\/cell-reports-s-laye-actualite\/_metadata\/content\/_data\/LarrieuT155-2.jpg_197x155\" alt=\"\" width=\"155\" height=\"197\" \/><\/span><strong>Thomas Larrieu (actuellement postdoc \u00e0 l\u2019EPFL, Lausanne, Suisse, dans le groupe du Pr. Laboratoire de g\u00e9n\u00e9tique comportementale (LGC).) :<\/strong> Une \u00e9tape est ainsi franchie dans la mise en lumi\u00e8re du substrat neurobiologique impliqu\u00e9 dans l\u2019adaptation au stress et l\u2019anxi\u00e9t\u00e9 qui se d\u00e9veloppe chez certains individus en situation de stress chronique. The relationship\u00a0between the CSDS model and a physiologically\u00a0pertinent paradigm of synaptic plasticity is not\u00a0known. Here, we found that cluster analysis of the\u00a0emotional behavior states of mice exposed to\u00a0CSDS allowed their segregation into anxious and\u00a0non-anxious groups. Endocannabinoid-mediated\u00a0spike-timing dependent plasticity (STDP) in the\u00a0nucleus accumbens was attenuated in non-anxious\u00a0mice and abolished in anxious mice. Anxiety-like\u00a0behavior in stressed animals was specifically correlated\u00a0with their ability to produce STDP. Pharmacological\u00a0enhancement of 2-arachidonoyl glycerol\u00a0(2-AG) signaling in the nucleus accumbens normalized\u00a0the anxious phenotype and STDP in anxious\u00a0mice.
