For over 20<\/span> <span dir=\"ltr\" role=\"presentation\">years it has <\/span><span dir=\"ltr\" role=\"presentation\">been thought that the CNS environment was the main cause of this lack of regeneration. <\/span><span dir=\"ltr\" role=\"presentation\">Indeed, after CNS injury, myelin debris release and glia scar formation act as a chemical <\/span><span dir=\"ltr\" role=\"presentation\">and physical barrier for <\/span><span dir=\"ltr\" role=\"presentation\">axon outgrowth. Surprisingly their modulation in vivo did not unlock <\/span><span dir=\"ltr\" role=\"presentation\">CNS repair. Since <\/span><span dir=\"ltr\" role=\"presentation\">2008, it has been shown that neurons themselves <\/span><span dir=\"ltr\" role=\"presentation\">loose<\/span> <span dir=\"ltr\" role=\"presentation\">the ability to <\/span><span dir=\"ltr\" role=\"presentation\">regenerate over the course of development. <\/span> <span dir=\"ltr\" role=\"presentation\">We showed that t<\/span><span dir=\"ltr\" role=\"presentation\">his capacity is further <\/span><span dir=\"ltr\" role=\"presentation\">decreased by the <\/span><span dir=\"ltr\" role=\"presentation\">injury itself. <\/span><span dir=\"ltr\" role=\"presentation\">Thus, <\/span><span dir=\"ltr\" role=\"presentation\">by <\/span><span dir=\"ltr\" role=\"presentation\">modulating the neuronal intrinsic molecular pathways <\/span><span dir=\"ltr\" role=\"presentation\">we are now able to induce long distance regeneration. However, another unexpected <\/span><span dir=\"ltr\" role=\"presentation\">roadblock stands on the way to achieve functional recovery: regenerating axons present <\/span><span dir=\"ltr\" role=\"presentation\">strong guida<\/span><span dir=\"ltr\" role=\"presentation\">nce defect<\/span><span dir=\"ltr\" role=\"presentation\">s<\/span> <span dir=\"ltr\" role=\"presentation\">that counteract any attempt to build a functional circuit.
