Abstract :

The seven-pass transmembrane cadherins Celsr1, Celsr2 and Celsr3 are the mammalian homologues of the Drosophila Flamingo, a key component of the planar cell polarity (PCP) signaling pathway. In Flies, the PCP governs the polarity of epithelial cells in a plane orthogonal to the apical-basal axis, and one feature of such a polarity is the stereotypic organization of the wing hairs. In the mammalian central nervous system, Celsr1 is expressed by neuron precursors, Celsr3 by postmigratory neurons, and Celsr2 by both (Tissir et al. 2002). To study the function of Celsr1-3 in vivo, we knocked out the three genes in mice. In this seminar, I will focus on the role of Celsr proteins in i) guiding axons of the internal capsule (Tissir et al. 2005; Zhou et al. 2008a; Zhou et al. 2008b), ii) the polarity of the ependymal layer lining the cerebral ventricles (unpublished), and iii) the tangential migration of facial bronchiomotor neurons from medial rhombomere 4 to lateral rhombomere 6 (unpublished).
